Pharmacology

Response fades fast over a few doses

Tachyphylaxis is a rapid fall in response to a drug after only a few doses given in quick succession, a fast-onset form of tolerance distinct from the slow tolerance that builds over weeks. Rang & Dale's pharmacology and Goodman & Gilman's the pharmacological basis of therapeutics describe mechanisms such as depletion of a releasable mediator, receptor desensitisation, or receptor internalisation that blunt the effect within minutes to hours. The referent is a steep, short-timescale loss of drug effect on repeated dosing, which can often be restored by a rest interval that lets the system recover.[1][2]

Unit
The decline in drug response over successive closely spaced doses
Recovery means
Response typically returns after a dosing pause that lets mediators replenish or receptors resensitise
Measured by
Observed as a falling effect across rapidly repeated equal doses in the same preparation or patient
