Molecular genetics

A DNA element that boosts transcription from afar

In molecular genetics, an enhancer is a regulatory DNA element that increases transcription of a target gene, acting over distance and in either orientation by binding activator proteins that loop to the promoter. Banerji, Rusconi, and Schaffner's 1981 Expression of a β-globin gene is enhanced by remote SV40 DNA sequences identified the first such element, showing that a remote sequence could raise expression regardless of position. The referent is a cis-acting control sequence defined by its distance- and orientation-independent boost to transcription, measured by changes in transcript level, and it is a different construct from a food additive or a permeation agent.[1]

Unit
The fold increase in transcription of the target gene attributable to the element
Recovery means
Not applicable
Measured by
Quantified by reporter-gene or transcript assays comparing expression with and without the element, across positions and orientations
