Genomics

sequencing DNA by breaking it into random fragments, reading each, and reassembling the order computationally from overlapping reads.

In genomics, shotgun sequencing breaks DNA into many random fragments, sequences them independently, and reconstructs the original order computationally by overlapping the reads, so coverage is sprayed across the genome rather than read end to end. Staden's 1979 strategy introduced the computational assembly approach, and Fleischmann and colleagues' 1995 whole-genome sequencing of Haemophilus influenzae proved it at the scale of a complete bacterial genome. The referent is a fragment-and-reassemble sequencing method, measured by read coverage and assembly contiguity, named for the scatter of a shotgun rather than for the firearm or a football formation.[1][2]
