Microbiology & immunity

a short DNA segment captured from an invader and stored in a CRISPR array, guiding Cas nucleases on re-infection.

In bacterial and archaeal CRISPR–Cas immunity, a spacer is a short segment of DNA captured from an invading phage or plasmid and stored between the repeat sequences of a CRISPR array, where it is transcribed into a guide that directs Cas nucleases to cut matching invader DNA on re-infection. Jansen and colleagues named the repeat–spacer loci in 2002, and Barrangou and colleagues showed in 2007 that the spacers are an adaptive, heritable immune memory. Unlike passive intergenic spacer DNA, these spacers are an active record of past infection, a distinct construct that merely shares the word.[1][2]
